Abstract

Aquesta tesi suposa la continuació en el desenvolupament d’una reacció de transmetal·lació [2+1] per la preparació de radiofàrmacs de segona generació. Breument, aquesta consisteix en la directe bioconjugació del fragment fac-{99mTc(CO)3}+ en condicions d’alta activitat específica emprant un precursor de zinc que conté una molècula biològicament activa. S’han sintetitzat dos nous complexos de reni mitjançant dos complexos ja descrits de zinc bioconjugays amb la glucosamina i la piperazine. A més, s’han usat dos lligands π-acceptors, trifenilfosfina trisulfonada i isocianur de terc-butil, per millorar les característiques del potencial radiotraçador, i, al mateix temps, l’ús de diferents lligands permet avaluar la versatilitat de la reacció de transmetal·lació [2+1]. D’altra banda, atès que l’àcid fòlic ha despertat gran interès per dirigir fàrmacs a cèl·lules tumorals, s’han sintetitzat dos nous compostos fr zinc bioconjugats amb aquesta biomolècula per ser aplicats en la reacció de transmetal·lació [2+1]. La preparació d’aquests complexos han permès l’obtenció dels corresponents complexos de reni. Tots els complexos de reni mencionats han servit com a model per caracteritzar els corresponents homòlegs de tecneci-99m sintetitzats mitjançant la reacció de transmetal·lació. L’èxit en l’obtenció dels quatre potencials radiofàrmacs ens permet afirmar que la reacció de transmetal·lació [2+1] és una estratègia apropiada i robusta per la bioconjugació del precursor fac-{99mTc(CO)3}+ donada la seva simplicitat experimental. A més a més, els radiotraçadors per aplicacions en càncer han estat estudiats in vivo i in vitro mostrant una clara activitat biològica. Finalment, un nou complex lipofílic de zinc ha estat sintetitzat i caracteritzat per poder ser aplicat en la mencionada reacció. Malgrat que aquest producte no s’incorpora en liposomes, la reacció de transmetal·lació dona lloc als complexos de reni o l’homòleg de tecneci-99m, el seu elevat caràcter amfifílic facilita la seva interaccionar amb els fosfolípids de les vesícules, de tal manera que la reacció permet, d’una manera simple i funcional, el radiomarcatge de vesícules preformades.


This thesis can be seen as a continuation of a previous work in our research group based on the development of a transmetallation reaction for the preparation of second generation radiopharmaceuticals. Briefly, the low solubility of the bioconjugated zinc complex allows the straightforward radiolabeling of fac-{99mTc(CO)3}+ core, with this method the final potential radiopharmaceutical with high specific activity. In this work, two new rhenium complexes have been synthetized by means of a reported zinc complexes derivatized with glucosamine and piperazine. Two π-acceptors ligands, triphenylphosphine trisulfonate and tert-butyl isocyanide, have been used to improve the features of the potential radiotracer, and, at the same time, the use of this ligands will test the versatility of the [2+1] transmetallation reaction. On the other hand, since in the last few years folic acid has awakened interest as a biovector, two new zinc complexes have been synthetized to be used in the transmetallation reaction. Afterwards, the corresponding rhenium complex has been synthetized as a model for the final technetium homologue. The proper obtention of the bioconjugated rhenium complexes has paved the way to perform the radiosynthesis with technetium-99m. After some optimizations, we are able to affirm that the [2+1] transmetallation reaction is a very convenient reaction to bioconjugate fac-{99mTc(CO)3}+ because of its simplicity from an experimental point of view. Moreover, the radiotracers bioconjugated with glucosamine and folic acid derivatives have been studied in vivo and in vitro. These studies have shown a clear biological activity for each potential radiopharamaceuticals. Finally, a lipophilic zinc complex has been synthetized and characterized to apply the above-mentioned reaction for liposome radiolabeling. Despite this product is not anchored in the phospholipid bilayer, the corresponding rhenium and technetium-99m products show an amphiphilic behavior. Thus, since amphiphilic molecules can interact with phospholipids, in this work is shown how the [2+1] transmetallation reaction can be useful for liposome radiolabeling.
